Is htaccess affecting my form posts?

For example, if I attempt to save the option under http://mysite.com/admin/layout it posts back to that same URL but as a page not found which is frustrating as it posts to itself and comes up fine when POST isn't set. Thanks in advance.

What is your htaccess? We cannot know if there is a problem without seeing it!
